Rnd1 is a small (~21 kDa) signaling G protein (more specifically a GTPase), and is a member of the Rnd subgroup of the Rho family of GTPases. It is encoded by the gene RND1. It contributes to regulating the organization of the actin cytoskeleton in response to extracellular growth factors (Nobes et al., 1998). GTPases (singular GTPase) are a large family of hydrolase enzymes that can bind and hydrolyze guanosine triphosphate (GTP). The GTP binding and hydrolysis takes place in the highly conserved G domain common to all GTPases. The hydrolysis of the ? phosphate of GTP into guanosine diphosphate (GDP) and Pi, inorganic phosphate, occurs by the SN2 mechanism (see nucleophilic substitution) via a pentavalent intermediate state and is dependent on the magnesium ion Mg2+.
